The sodium iodide symporter and thyroid disease.

چکیده
The movement of a variety of species from the iodine-rich environment of the sea to the relatively iodine-deficient land has necessitated the development of mechanisms which will trap more efficiently inorganic iodide (I – ) necessary for the formation of thyroid hormones (Venturi et al. , 2000). In man as in other mammals iodide uptake by the thyroid is mediated via a TSHdependent transmembrane protein, the sodium iodide symporter (NIS), so-called because it co-transports Na + with I – into the thyroid against the concentration gradient (De La Vieja et al. , 2000). The NIS is located in the basolateral membrane of thyroid follicular cells, and accumulated I – is organified to molecular iodine (I 2 ) through the action of the enzyme thyroid peroxidase (TPO) in the presence of H 2 O 2 , which takes place mainly at the apical membrane of the follicular cell. Organified iodine iodinates the thyroid protein thyroglobulin, which when stored in the lumen of thyroid follicles represents a storage site for the hormones thyroxine (T4) and triiodothyronine (T3) (Taurog, 1996). Possession of NIS enables the thyroid to concentrate I –

Differential regulation of the human sodium/iodide symporter gene promoter in papillary thyroid carcinoma cell lines and normal thyroid cells.
The absence of TSH-stimulated radioiodide uptake in differentiated thyroid cancer is associated with a high recurrence rate and reduced survival. We studied regulation of the sodium/iodide symporter gene in human papillary thyroid cancer cell lines (BHP) and primary human thyroid cells. BHP cells expressed very low levels of sodium/iodide symporter mRNA and did not concentrate iodide, but iodid...
متن کاملMethylation of sodium iodide symporter promoter correlated with aggressiveness and metastasis in papillary thyroid carcinoma: a meta-analysis.
BACKGROUND Methylation of sodium iodide symporter promoter has been reported to increase the incidence of papillary thyroid carcinoma (PTC). In this meta-analysis stratified via methylation of sodium iodide symporter promoter, we evaluate the relationship between methylation of sodium iodide symporter promoter and PTC. The association between methylation with aggressiveness and metastasis poten...
متن کاملLack of correlation for sodium iodide symporter mRNA and protein expression and analysis of sodium iodide symporter promoter methylation in benign cold thyroid nodules.
Cold thyroid nodules (CTNs) are characterized by a reduced iodide uptake in comparison to normal thyroid tissue. The sodium iodide symporter (NIS) is the first step in thyroid hormone synthesis and mediates the active iodide transport in the thyroid cells suggesting that decreased iodide uptake could be a result of changes in NIS expression or molecular defects in the NIS gene. In contrast to p...
متن کاملResveratrol Inhibits Sodium/Iodide Symporter Gene Expression and Function in Rat Thyroid Cells
Resveratrol is a polyphenol found in grapes and berries that has antioxidant, antiproliferative and anti-inflammatory properties. For these reasons, it is available as a dietary supplement, and it is under investigation in several clinical trials. Few data are available regarding the effects of resveratrol on thyroid function. A previous study showed that resveratrol transiently increases iodid...
متن کاملPendrin and sodium/iodide symporter protein expression in the testicular tissue of normal and diabetic rats in prepubertal and post pubertal stages
Pendrin (PDS) and sodium/iodide symporter (NIS) are transmembrane proteins that are located in numerous tissue types, particularly thyroid follicular epithelial cells, where they are entrusted with the regulation of iodine molecules. In the present study, we aimed to clarify changes in PDS and NIS protein expression, in the testicular tissue of prepubertal and <span la...
